Ce sont les premi\u00e8res cellules sanguines \u00e0 migrer vers le site d'infection o\u00f9 elles lib\u00e8rent de grandes quantit\u00e9s d'esp\u00e8ces r\u00e9actives de l'oxyg\u00e8ne (ROS) et plusieurs peptides et enzymes n\u00e9cessaires \u00e0 la destruction microbienne. Cependant, une activation excessive des neutrophiles peut induire des l\u00e9sions tissulaires participant \u00e0 l'inflammation, ainsi la caract\u00e9risation des enzymes impliqu\u00e9es dans l'activation des neutrophiles pourrait aider \u00e0 identifier de nouvelles cibles pharmacologiques pour traiter l'inflammation. La prolyl-isom\u00e9rase Pin1 est une enzyme ubiquitaire impliqu\u00e9e dans plusieurs fonctions, cependant, son r\u00f4le dans les fonctions des neutrophiles est moins connu. Dans cette \u00e9tude, nous montrons que le peptide bact\u00e9rien N-formyl-m\u00e9thionyl-leucyl-ph\u00e9nylalanine (fMLP ou fMLF), un agoniste du r\u00e9cepteur coupl\u00e9 aux prot\u00e9ines G (RCPG) induit l'activation de Pin1 dans les neutrophiles humains. PiB et juglone, deux inhibiteurs de Pin1 ont inhib\u00e9 l'activit\u00e9 de Pin1 dans les neutrophiles et, par cons\u00e9quent, inhib\u00e9 la chimiotaxie induite par la fMLP et la d\u00e9granulation de l'azurophile et des granules sp\u00e9cifiques, mesur\u00e9es respectivement par la my\u00e9loperoxydase et la lib\u00e9ration de lipocaline associ\u00e9e \u00e0 la g\u00e9latinase des neutrophiles (NGAL). Nous avons \u00e9galement montr\u00e9 que PiB inhibait la production de superoxyde induite par le TNF\u03b1 + fMLP, confirmant l'effet de la juglone. Ces donn\u00e9es montrent que les inhibiteurs de Pin1 alt\u00e8rent les fonctions cl\u00e9s des neutrophiles pro-inflammatoires provoqu\u00e9es par l'activation du GPCR et sugg\u00e8rent que Pin1 pourrait contr\u00f4ler les fonctions inflammatoires des neutrophiles.<\/p>[:]","paragraphe_en":"[:fr]<h2 class=\"title\" style=\"text-align: justify;\">Abstract<\/h2>\r\n<div id=\"enc-abstract\" class=\"abstract-content selected\">\r\n<p style=\"text-align: justify;\">Neutrophils are key cells of the innate immune and inflammatory responses. They are the first blood cells to migrate to the infection site where they release high amounts of reactive oxygen species (ROS) and several peptides and enzymes required for microbial killing. However, excessive neutrophil activation can induce tissue injury participating in inflammation, thus the characterization of the enzymes involved in neutrophil activation could help to identify new pharmacological targets to treat inflammation. The prolyl-isomerase Pin1 is a ubiquitous enzyme involved in several functions, however, its role in neutrophil functions is less known. In this study, we show that the bacterial peptide N-formyl-methionyl-leucyl-phenylalanine (fMLP or fMLF), a G-protein coupled receptor (GPCR) agonist-induced Pin1 activation in human neutrophils. PiB and juglone, two Pin1 inhibitors inhibited Pin1 activity in neutrophils and consequently inhibited fMLP-induced chemotaxis and -degranulation of azurophil and specific granules as measured by myeloperoxidase and neutrophil gelatinase-associated lipocalin (NGAL) release respectively. We also showed that PiB inhibited TNF\u03b1 + fMLP-induced superoxide production, confirming the effect of juglone.
