class=\"sub-title\"><span>Contexte et objectifs:\u00a0<\/span><\/strong><span>Les mutations du domaine 2 [NOD2] d'oligom\u00e9risation des nucl\u00e9otides sont des facteurs de risque cl\u00e9s de la maladie de Crohn [MC].\u00a0NOD2 contribue \u00e0 l'hom\u00e9ostasie intestinale en r\u00e9gulant l'immunit\u00e9 inn\u00e9e et adaptative ainsi que la fonction \u00e9pith\u00e9liale intestinale.\u00a0Cependant, les r\u00f4les exacts de NOD2 dans la MC et d'autres troubles associ\u00e9s \u00e0 NOD2 restent mal connus.<\/span><\/p>\r\n<p style=\"text-align: justify;\"><strong class=\"sub-title\"><span>M\u00e9thodes:\u00a0<\/span><\/strong><span>Nous avons initialement observ\u00e9 que l'expression de NOD2 \u00e9tait augment\u00e9e dans les cellules \u00e9pith\u00e9liales loin des zones enflamm\u00e9es chez les patients atteints de CD.\u00a0Pour explorer cette d\u00e9couverte, l'expression de l'ARNm Nod2, l'inflammation et l'expression des cytokines ont \u00e9t\u00e9 examin\u00e9es dans l'intestin gr\u00eale de souris de type sauvage [WT], Nod2 knockout et mutantes Nod2 apr\u00e8s instillation rectale d'acide 2,4,6-trinitrobenz\u00e8ne sulfonique [TNBS] .<\/span><\/p>\r\n<p style=\"text-align: justify;\"><strong class=\"sub-title\"><span>R\u00e9sultats:\u00a0<\/span><\/strong><span>Chez les souris WT, la r\u00e9gulation ascendante de Nod2 en amont de la l\u00e9sion rectale \u00e9tait associ\u00e9e \u00e0 une expression pro-inflammatoire de cytokines, mais pas de l\u00e9sions inflammatoires histologiques manifestes.\u00a0Inversement, chez les souris d\u00e9ficientes en Nod2, l'inflammation s'est propag\u00e9e de la colite \u00e0 l'il\u00e9on et au duod\u00e9num.<\/span><\/p>\r\n<p style=\"text-align: justify;\"><strong class=\"sub-title\"><span>Conclusions:\u00a0<\/span><\/strong><span>Nod2 prot\u00e8ge l'intestin de la colite qui se propage \u00e0 l'intestin However, the exact roles of NOD2 in CD and other NOD2-associated disorders remain poorly known.<\/p>\r\n<p><strong class=\"sub-title\">Methods:\u00a0<\/strong>We initially observed that NOD2 expression was increased in epithelial cells away from inflamed areas in CD patients. To explore this finding, Nod2 mRNA expression, inflammation, and cytokines expression were examined in the small bowel of wild-type [WT], Nod2 knockout and Nod2 mutant mice after rectal instillation of 2,4,6-trinitrobenzene sulphonic acid [TNBS].<\/p>\r\n<p><strong class=\"sub-title\">Results:\u00a0<\/strong>In WT mice, Nod2 upregulation upstream to rectal injury was associated with pro-inflammatory cytokine expression but no overt histological inflammatory lesions.
