The aim of the study was to characterize the molecular heterogeneity of CCs by a global proteomic approach.<\/p>\r\n<h4 style=\"text-align: justify;\">EXPERIMENTAL DESIGN:<\/h4>\r\n<p style=\"text-align: justify;\">Thirty-three tumor samples from 17 intrahepatic CCs (iCC) (9 developed on normal (iCC<sub>N<\/sub>\u00a0) and 8 developed in cirrhotic liver (iCC<sub>C<\/sub>\u00a0)); 5 hilar CCs (CC<sub>H<\/sub>\u00a0); 5 pancreatic CCs (CC<sub>P<\/sub>\u00a0) and 6 hepatocellular carcinomas (HCC), were submitted to label-free quantitative proteomic analysis. Differential proteins were analyzed by immunohistochemistry in a validation set of 30 CCs.<\/p>\r\n<h4 style=\"text-align: justify;\">RESULTS:<\/h4>\r\n<p style=\"text-align: justify;\">Unsupervised analysis revealed two main clusters: cluster 1 contained most of the iCC<sub>C<\/sub>\u00a0while cluster 2 was divided in 2 subgroups, one containing most of the iCC<sub>N<\/sub>\u00a0and the other regrouping CC<sub>H<\/sub>\u00a0and CC<sub>P<\/sub>\u00a0. Compared to iCC<sub>N<\/sub>\u00a0, iCC<sub>C<\/sub>\u00a0displayed upregulation of molecules involved in cell adhesion, motility and angiogenesis. Epithelial markers associated with secretory pathway and fibroblast markers were overexpressed in CC<sub>H<\/sub>\u00a0compared to iCC<sub>N<\/sub>\u00a0CONCLUSION AND CLINICAL RELEVANCE: This study demonstrated that iCC<sub>C<\/sub>\u00a0is a specific entity, suggesting a major impact of the background liver on tumor biology, and confirmed that extrahepatic CCs define a homogeneous subgroup.<\/p>\r\n<\/div>","paragraphe_en":"","documents":null},"yoast_head":"<!-- This site is optimized with the Yoast SEO plugin v27.4 -
